João was one of the lords taken prisoner at the Battle of Alcácer Quibir and ransomed, along with his brother Luís.

Son of Luís de Lencastre and Magdalena of Granada, he was married firstly to Paula da Silva, with issue:
• Luís
• Lourenço, married to Ignez de Noronha (Telles de Meneses) (†1651), with issue
• Jorge
• Catharina, married to Fernaõ Martins Mascarenhas, Senhor de Lavre, with issue

João was married secondly to Filipa de Castro, with no issue
